The Guide System is a new feature in Shadowlands where veteran players who are willing to help new players can do so more easily, via the Newcomer Chat. In this post, we went through the Guide system from both perspectives!
Becoming a Guide
Players interested in becoming a Guide can do so by talking to the Guide Recruiter NPCs - Alliance players can talk to Celeste Periwinkle by the Stormwind Embassy and Horde players can talk to Eyla Pathleader by the Orgrimmar Embassy.
There are specific requirements to become a Guide. Currently, these involve completing several achievements in Battle for Azeroth as well as reaching level 50, but expect these to be updated at some point in Shadowlands.

Once you become a Guide, you will gain access to the Newcomer Chat, with the default chat bind being /4.
Newcomer Chat
The main feature of the Guide system is the newcomer chat, a separate chat that appears anywhere in the world where guides and newcomers interact. New WoW players with Starter Accounts will have access immediately to the Newcomer chat when logging into the game (once again, using the default /4 chat bind to gain access to the chat):
In the Newcomer Chat, newcomers are indicated by a
Murloc icon
and guides are indicated by a
Green Flag
(as well as a Guide) text indicator next to their names. Newcomers won't see the murloc icons, and guides won't see the green flag icons.

Guide and Newcomer World Icon Indicators
Besides their chat indicators in the Newcomer chat, Guides and Newcomers can identify each other in the world - Guides will have green flags above their head, while Newcomers will have murlocs.
Only guides can see murloc icons, and only newcomers can see the green flags.
Leaving the Guide System
If you are a guide and you no longer wish to be one, you can simply return to either Guide Recruiter NPCs in your faction and talk to them to leave the system. There is no punishment for leaving the system, and you can return at any time!
It is currently unknown how long until newcomers are able to leave the Newcomer chat, but presumably they will be able to do so once they purchase a Shadowlands license to their WoW account.
